Mine leaked from the plastic baffle at the bottem of door pillar get it all dry and drive car for a week or more when its rained keep Checking the floor pan i had to use a very good auto mate mastic Not silocone you will then find the leaking

vent leak
hi its the same place as mine was leaking from then must be a few cars out there with this problem get a good quality mastic and don't put anything back till its 100% cured the leak

Its just a blanking plate with a rubber seal, im not going to pull it off in case it makes things worse, i will seal it with roofing sealant.

If its a blanking seal/cover and the water is behind it then the water must be coming in somewhere. If you seal it up then the water will still be coming in and will find somewhere else to go to. I agree with jpliddy. Remove it and have a look behind it.
